Identifying the Telomerase RNA of Aspergillus nidulans: the generation of a DNA construct and refinement of transformation procedures
نویسنده
چکیده
Telomeres are genomic structures found at the ends of linear chromosomes that protect cells by ensuring the complete transmission of genetic information to future generations. As the cell replicates, the telomeres will naturally shorten, allowing them to be used as a biological clock. The enzyme telomerase, a complex molecule made of a protein (TERT) and an RNA component (TR), regulates telomere length. The TR region of the enzyme contains the template region used to synthesize the telomeric repeats, TTAGGG, which are found in most mammals as well as in our model organism of choice — Aspergillus nidulans. In this project, I attempt to knockout a putative TR template. Fusion PCR was used to synthesize a DNA construct that was transformed into fungal cells and integrated into the genome through homologous recombination. I hypothesize that this putative TR template mutation will result in an abnormal phenotype in the fungal cells that arises from the shortening of telomeres due to the dysfunctional telomerase.

Down-Regulation of sidB Gene by Use of RNA Interference in Aspergillus nidulans
Background: Introduction of the RNA interference (RNAi) machinery has guided the researchers to discover the function of essential vital or virulence factor genes in the microorganisms such as fungi. In the filamentous fungus Aspergillus nidulans, the gene sidB plays an essential role in septation, conidiation and vegetative hyphal growth. In the present study, we benefited from the RNAi strate...

Background: Aspergillus species are occasionally associated with human infections. It is essential to find a rapid and reliable method to identify these fungi and can be useful for screening the disease and also epidemiological aims. Materials and Methods: Our subjects included Aspergillus species isolated from clinical and environmental sources of four Iranian educational hospitals. Aspergilli...
متن کاملNovel Telomere-Anchored PCR Approach for Studying Sexual Stage Telomeres in Aspergillus nidulans
Telomere length varies between germline and somatic cells of the same organism, leading to the hypothesis that telomeres are lengthened during meiosis. However, little is known about the meiotic telomere length in many organisms. In the filamentous fungus Aspergillus nidulans, the telomere lengths in hyphae and asexual spores are invariant. No study using existing techniques has determined the ...
متن کاملA Robust Analytical Pipeline for Genome-Wide Identification of the Genes Regulated by a Transcription Factor: Combinatorial Analysis Performed Using gSELEX-Seq and RNA-Seq
For identifying the genes that are regulated by a transcription factor (TF), we have established an analytical pipeline that combines genomic systematic evolution of ligands by exponential enrichment (gSELEX)-Seq and RNA-Seq. Here, SELEX was used to select DNA fragments from an Aspergillus nidulans genomic library that bound specifically to AmyR, a TF from A. nidulans. High-throughput sequencin...
متن کاملTransformation of Aspergillus nidulans by microprojectile bombardment on intact conidia.
This paper describes transformation of intact conidia of Aspergillus nidulans, auxotrophic for arginine, by using the biolistic process. The plasmid employed was pFB39, carrying the argB gene. The transformation frequency obtained was 81 transformants/microgram of DNA. Classical genetics and molecular analysis were conducted to analyse transformants and to determine in which chromosome integrat...
